Provided is a method for preparing human mesenchymal stem cells capable of delaying cellular senescence and resisting malignant transformation. The preparation method is to mutate the codon encoding the 253rd amino acid residue of the FOXO3 protein from the codon encoding serine to the codon encoding alanin, and the codon encoding the 315th amino acid residue of the FOXO3 protein. It involves making two mutations to the pluripotent stem cell genome, with a mutation from a serine-encoding codon to an alanin-encoding codon. Recombinant mesenchymal stem cells prepared by the above method, and their use in cell transplantation therapy are further provided. [Selection diagram] None
