摘要
Disclosed is a bioartificial liver based on human iPSCs-induced hepatocyte-like cells and a multilayer porous bioreactor (14), the bioartificial liver comprising: a plasma separation/transfusion circulation part comprising a blood input tube (1), a gas discharge tube spring clamp (7), a blood input peristaltic pump (2), a heparin pump (3), a plasma separation column (4), a first pressure monitor (5) and a heater (6), which are connected in sequence; and a cell reactor/plasma component exchange double-circulation part comprising a plasma input peristaltic pump (8) sequentially connected to a plasma outlet (4-2), and a semipermeable membrane exchange column (10), a plasma exchange peristaltic pump (11), a red blood cell pool (12), a membrane oxygenation unit (13), a multilayer porous bioreactor (14), a second pressure detector (15) and a third pressure monitor (16) which are arranged in a special incubator (9) having a temperature of 37℃, with an outlet of the third pressure monitor (16) and a blood cell outlet (4-3) being connected together to an inlet of the first pressure monitor (5) and then sequentially connected to the heater (6) and a blood output tube (17). The cooperation of the two circulation parts can maintain the temperature and oxygen supply required by cells and also improve the liquid exchange efficiency and biological compatibility, meets clinical requirements, and can also reduce the risk of heterogeneous viral infection and the risk of an immunological reaction.
