Human pluripotent stem cells (hPSCs) have dual value as microphysiological experimental models and regenerative therapeutics. Although hPSCs are epithelial cells, little is known to what extent hPSCs and progeny epithelia can reconstitute lineage-specific functions. Here, we show that hPSCs and their differentiated progeny in three-dimensional culture can functionally reproduce tissue-specific epithelial morphogenesis, physiology, and disease.
