PROBLEM TO BE SOLVED: To provide a method for overcoming a disadvantage in that, in the silkworm belonging to the family Bombycidae, the back silk gland is thin and difficult to extract and the middle silk gland has a low synthesis amount.;SOLUTION: According to the present invention, unlike the silkworm, the attention has been focused on the silk glands of Eri silkworm belonging to the family Antheraea. It is found that the silk gland of this Eri silkworm rapidly develops at the fifth instar stage and furthermore it can be easily removed entirely with almost no distinction between the middle and back silk glands. Using the easily extracted silk glands, the conditions for extract production has been investigated in order to improve the synthesis amount to a practical level. As a result, a cell-free protein synthesis system with a synthesis amount of 4.2 μg/ml has been successfully constructed using the silk gland removed from the fifth instar Eri silkworm larvae at day 2 or 3 and using an extract prepared by adding an extraction liquid 1.25 times greater than the weight of the silk gland. As a result, it is possible to produce a silk gland extract solution with high synthetic ability while maintaining improved workability.
