The present invention relates to a primer for pyrosequencing for early diagnosis of mad cow disease by detecting a somatic mutation associated with mad cow disease in a bovine prion protein gene. By using the composition, kit, and detection method for detecting mad cow disease-associated somatic mutation according to the present invention, it is possible to economically and accurately detect an individual having an extremely low distribution of mad cow disease-related somatic mutation. Therefore, it is possible to contribute to pre-existing disaster prevention of mad cow disease by selecting individuals highly susceptible to mad cow disease at an early stage.
