1342391 Thymocytary antigens and antiserum ALBERT ROLLAND SA 2 Feb 1972 [9 Feb 1971] 4276/71 Heading ASB Animal thymocytary antigens are prepared and preserved by (a) forming a cellular suspension of thymocytes from an animal thymus or from the Fabricius bursa of a bird, (b) filtering the suspension on a series of increasingly fine screens or on a number of layers of gauze, (c) centrifuging the filtrate, (d) resuspending the sediment in buffered physiological solution, (e) destroying the erythrocytes, (f) washing the sediment with buffered physiological solution with centrifuging, (g) checking the cell suspension so obtained for the absence of cellular contaminants, (h) freezing and preserving the suspension at -20 to -40‹C and (i) when required for use, thawing the suspension and dissociating it by agitation in the presence of spherical balls. An antithymocytary serum specific to one animal species may be prepared by (a) injecting the antigen product of this process into an animal of a different species, (b) taking serum from the animal during immunization and testing it for soluble circulating antigent-antibody complexes, (c) harvesting serum from the immunized animal and (d) purifying the serum by adsorption on a polymer prepared from glutaraldehyde and protein derived from the liver and serum of the animal from which the antigen was obtained or another animal of the same species.
