The arrangement is intended for sepg. solutes from a mixt. by radial thin-layer chromatography. The chromatography plate carrying the absorbent is rotated during or after the central application of the eluent (washing agent). The plate is preferably rotated about its centre at a speed of 100-6000 rpm (usually 1500-3000 rpm). The eluent may be supplied under pressure. The plate is pref. clamped between a (driven) carrier and a cover plate provided with a layer of sealing material (3) such as PTFE. Densitometric quantitative analysis may be carried out by rotating the plate using an optical scanning detector linearly moved across it. Used in radial thin-layer chromatographic separator partic. for qualitative and/or quantitative analysis, e.g. of metabolites. Arrangement overcomes a range of drawbacks described in some detail and experienced with standard, high-performance, and overpressurised thin-layer chromatography (TLC, HPTLC, OPTLC). By increasing rotational speed, the time for analysis is considerably reduced (for a quoted example from 90 min., using TLC, to 6 min) whilst the quality of sepn. is optimised. Smaller absorbent particles can be used without increasing the analysis time which increases the separation power. Colouring agents can be applied more homogeneously than with a non-rotating plate when spray-on methods are used. Arrangement enables applying a large quantity of a mixt. contg. one or more very low-concn. components, e.g. under vacuum by a syringe on to the rotating plate, to obtain satisfactory analysis.
