AN IMPROVED METHOD OF CLONING DOUBLE-STRANDED RNA, AND ITS USE IN THE PRODUCTION OF VIRAL GENE CLONES

A method of direct insertion of double stranded RNA into a cloning vector formed of double-stranded DNA, comprising a) isolation of dsRNA from a source of interest; b) if necessary, capping the dsRNA; and c) the direct ligation of dsDNA dsRNA in the appropriate cloning vector. This method allows direct cloning of dsRNA, thereby eliminating the traditional step of firstly a copy of DNA complementary to the RNA molecule. Thanks to this method, it is possible to construct probes for the detection of viral infections in plants and animals. This method can also be used to produce new plant strains resistant to a particular virus, or to give the animals immunity to a specific virus.
