Process for the production of sucrose phosphorylase, sucrose phosphorylase obtained by the process and application to the synthesis of glucose-1-phosphoric acid and its salts. According to a preferred embodiment of the invention, the process consists in inoculating, with a strain of microorganism Leuconostoc mesenteroides classified ATCC 12291, a culture medium containing sucrose, tryptone, yeast extract, sodium or potassium phosphate, trace elements in solution, an antifoam comprising at least one ester of a polyether polyol and an unsaturated or polyunsaturated fatty acid. This process permits an industrial production of the enzyme. The subject of the invention is also the manufacture of glucose-1-phosphoric acid and its salts by bringing the enzyme obtained into contact with sucrose and at least one alkali metal phosphate salt.
