Glyco haemoglobin measurement by HPLC - by simultaneously sepg. glyco haemoglobin, haemoglobin and haemoglobin derivs. and removing labile haemoglobin to measure stable haemoglobin
Measurement (HPLC) comprises simultaneously sepg. glycohaemoglobin, haemoglobin and haemoglobin derivs. in blood process(es) and degrading haemoglobin in labile form in blood into glucose and haemoglobin removing the haemoglobin component in labile form (process (b)) and measuring the haemoglobin in stable form. USE/ADVANTAGE - Amt. of stable haemoglobin in blood has a good correlation with the blood glucose level of the last 1-3 months, but as stable haemoglobin and labile haemoglobin have similar structures, they are difficult to separate by liq. chromatography. Stable haemoglobin is rapidly sepd. from other haemoglobin components and inhibits the blocking up of pipes and filters. The process is rapid and reproducible and analyses stable haemoglobin simply without being affected by labile haemoglobin, the level of which varies with physiological factors.
