PURPOSE: To efficiently creat a low temperature resistant rice plant body capable providing low temperature resistance in the young plant body by subjecting calluses of rice to low temperature treatment, selecting a low temperature resistant callus and then redifferentiating rice young plant from this callus. ;CONSTITUTION: Rice calluses are treated with a cell wall-decomposing enzyme to prepare a protoplast and the protoplast is irradiated with ultraviolet rays from a distance of 30cm for 60 sec to induce variation and the variant is amhedded in an agar medium and cultured under dark conditions at 28°C for 10-40 days to form a micro colony. Then, the colony is moved to a medium containing a fatty acid-unsaturating enzyme inhibitor such as 4- chloro-5-(dimethylamino)-2-phenyl-3 (2H)-pridazinone together with agar piece and cultured at 28°C and 2000 1ux for 10 day and then subjected to low temperature treatment at 5°C under dark conditions for 30 day and again cultured at 28°C and 2000 lux and low temperature resistant viable callus is selected and young rice plant is redifferentiated from this callus to creat the objective low-temperature resistant rice plant body.;COPYRIGHT: (C)1994,JPO&Japio
