DETOXIFIED LPS-CHOLERA TOXIN CONJUGATE VACCINE FOR PREVENTION OF CHOLERA

The invention describes a vaccine formulation comprising conjugates composed of detoxified LPS and proteins comprising cholera toxin (CT). Treatment with hydrazine (DeA-LPS) reduces the endotoxic characteristics of LPS to acceptable levels, and produces a larger molecule, and with increased antigenic / activity, compared to the saccharide produced by acid hydrolysis. Conjugates using V. cholerae toxin are also described, which have low levels of pyrogens, no toxic activity compared to Chinese hamster ovary cells, and induce reinforcing responses in vibriocidal antibodies and antibodies against the toxin cholera when injected subcutaneously as saline in mice. Conjugates produced as a cholera vaccine induce the formation of the same antibodies as parenteral injection cell vaccines, but have improved immunological characteristics and safety.
