PURPOSE: To obtain an antibody useful for e.g. diagnosing or treating chronic arthrorheumatism because of being capable of easily, quickly and accurately assaying an antigen, i.e., a synthetic peptide having an amino acid sequence specific to a human phospholipase A2-activated protein. ;CONSTITUTION: Firstly, a peptide having an amino acid sequence of the formula and specific to a human phospholipase A2-activated protein (PLAP) is synthesized, and cysteine residue is introduced into the C-terminal of this peptide followed by binding the resultant peptide to e.g. maleimide-modified Tachypleus tridentatus hemocyanin (KLH) to prepare an antigen. Second, this antigen, together with an adjuvant, is administered to BALB/C mice, and after the final immunization, the splenocytes are taken and fused with myeloma cells, and the resultant hybridoma are cultured in a HAT selective medium to select the clone capable of producing the aimed antibody, and this clone is then cloned by limiting dilution analysis; the monoclonal hybridoma produced is then cultured, thus obtaining the aimed new anti-PLAP monoclonal antibody useful for e.g. PLAP-as saying reagents.;COPYRIGHT: (C)1995,JPO
