PURPOSE: To obtain a glucagon decomposition enzyme having action to decompose glucagon and vasoactive intestinal peptide and useful for the treatment, etc., of diseases caused by the presence of excessive glucagon and vasoactive intestinal peptide. ;CONSTITUTION: This glucagon decomposition enzyme is obtained by culturing tumor cells originated from human pancreatic cancer tissue and has the following enzymological properties. (1) It hydrolyzes the peptide bond shown in the formula by an arrow by using glucagon, vasoactive intestinal peptide and secretin as substrates; (2) isoelectric point is pI 6.8; (3) molecular weight is 83 kDa (SDS electrophoresis); (4) enzymatic reaction is inhibited by 1M antipain and is not inhibited by 1M leupeptin and 0.1mM pepstatln; (5) it has peptide fragments SNSTSYVK, YYASTSYDDTYK, NLTEEYDVSEGEIELLYEK, LYWFLDEAK and ELREVETYYDLLFEK.;COPYRIGHT: (C)1996,JPO
