Vectors comprising a nucleotide sequence of formula (I) or (II), are new. R-X-A-X-IRES-X-B-X-polyA (I) R-X-B-X-IRES-X-C-X-polyA (II) R is a regulatory sequence for constitutive or inducible gene expression; A is a gene coding for a protein capable of inducing tumorigenic growth of cells; IRES is a nucleotide sequence of cellular or synthetic origin that is responsible for internal initiation in the translation process Äinternal ribosomal entry siteÜ; B is a reporter gene encoding a detectable secreted protein; polyA is a nucleotide sequence for transcript poly-adenylation; C is a marker gene encoding a protein permitting resistance-based selection, and X is an optional linker sequence. Also claimed is a mammalian cell line stably transfected with a vector as described above.
