PROBLEM TO BE SOLVED: To obtain clone of Zooxanthella exhibiting high infection ratio to Zooxanthella-free Paramecium bursaria and elucidate infection process of cell ecology by treating Paramecium bursaria under specific conditions in a liquid in which a vegetable is immersed. ;SOLUTION: This method for cloning Zooxanthella comprises (i) treating Paramecium bursaria with ultrasonic wave which breaks Paramecium bursaria, but does not break Zooxanthella in a liquid in which vegetable such as lettuce is immersed, (ii) subjecting the liquid treated with ultrasonic wave to centrifugal separation treatment, (iii) washing the separated Zooxanthella suspension with culture solution, (iv) inoculating the Zooxanthella suspension into an agar medium containing a culture solution, (v) culturing the alga, preferably at 24±1°C for ≥30 day under illumination, (vi) passing through a range of operation comprising a step arbitrarily selecting one colony from colonies formed on an agar medium and scraping the colony, transferring the colony into the agar medium containing same culture medium and culturing the alga and then repeating steps (iv) to (vi) in plural times.;COPYRIGHT: (C)1998,JPO
