PROBLEM TO BE SOLVED: To obtain a microsatellite marker which is located in the vicinity of a semi-dwarf gene, has a specific microsatellite and flanking DNAs having specific sizes in both sides of the gene, allows simple and accurate determination of a semi-dwarf gene, and is useful, for example, to administrate purity of rice seed. ;SOLUTION: This marker has a microsatellite which is located in the vicinity within 0.5 cent Morgan(cM) a rice semi-dwarf gene (sd-1), consists of (GT/CA) two-base repeats, and a sequence at position 296-330 shown by formula I of a sequence at position 294-316 shown by formula II, and flanking DNAs having 15 bases or more in both sides of it. It is preferable to determine the presence or absence of semi-dwarf gene(s) by carrying out PCR using primers of a pair of microsatellite markers having a base sequence having continuous 15-40 bases arbitrarily selected from a sequence at position 1-295 shown by formula I and a sequence at position 331-506 shown by formula I, and a rice DNA as a template, detecting difference in length of amplified DNA fragments, followed by discriminating difference in repeating sequences in the marker.;COPYRIGHT: (C)1999,JPO
