PROBLEM TO BE SOLVED: To inhibit hydrophobic adsorption of lipoprotein to a packing material by using the buffer including the salt of monovalent anion and/or nonion surfectant, and having pH above a predetermined values as the eluant. SOLUTION: A sample to be injected from a sample injection device 5 is introduced into a separation column 6 with the eluant 1 sent by an eluant feeding pump 3, and separated by various lipoprotein in serum. The separated lipoprotein is mixed with the reaction reagent 2-1 and 2-2 with cholesterol sent by an emzyme liquid feeding pump to be reacted with a reaction coil 8. The reaction product is detected by an ultraviolet visible detector 9, and cholesterol in the lipoprotein is quantified by an analytical curve method. The eluant is a buffer of pH 6.0-9.0, preferably 7.0-8.0. By adding the salt of monovalent anion and/or nonion surfectant to the eluant, the hydrophobic adsorption of lipoprotein to the packing material can be inhibited.
