Detection of IgA, IgG and/or IgM antibodies to Mycoplasma pneumoniae in human serum or plasma samples comprises contacting sample with M. pneumoniae protein antigens immobilized on test strips, washing the strips, incubating the strips with enzyme labeled anti-IgA, anti-IgG and/or anti-IgM antibodies, washing the strips and adding a chromogenic/chemiluminescent substrate. Detection of IgA, IgG and/or IgM antibodies to Mycoplasma pneumoniae in human serum or plasma samples comprises: (a) contacting the sample with M. pneumoniae protein antigens that have been separated and immobilized on a set of test strips; (b) washing the test strips to remove unbound antibodies; (c) incubating the test strips with a combination of enzyme-labeled nonhuman anti-IgA, anti-IgG and/or anti-IgM antibodies; (d) washing the test strips to remove unbound labeled antibodies; and (e) adding a chromogenic or chemiluminescent substrate, whereby the set of test strips displays a pattern of colored or chemiluminescent bands characteristic of M. pneumoniae. An Independent claim is also included for a test kit for detecting IgA, IgG and/or IgM antibodies to M. pneumoniae in human serum or plasma samples, comprising: (a) test strips bearing separated and immobilized M. pneumoniae protein antigens; (b) enzyme-labeled nonhuman anti-IgA antibodies; (c) enzyme-labeled nonhuman anti-IgG antibodies; (d) a solution of a chromogenic or chemiluminescent substrate; and (e) a representation of a colored or chemiluminescent band pattern typical of a positive M. pneumoniae assay.
