METHOD FOR DETECTING MULTIPLE DNA BINDING PROTEIN AND DNA INTERACTIONS IN A SAMPLE, AND DEVICES, SYSTEMS AND KITS FOR PRACTICING THE SAME

Methods for detecting the presence of at least one, usually a plurality of,DNA binding proteins, e.g., transcription factors, in a sample, bothqualitatively and quantitatively, are provided. In the subject methods, asubstrate having one or more DNA probes immobilized on a surface thereof, onefor each DNA binding protein of interest, is contacted with a sample underconditions sufficient for binding complexes of the probes and their respectiveDNA binding proteins to be produced. The sample may be purified with respectto one or more DNA binding proteins or be a cellular/nuclear extract.Resultant binding complexes on the surface of the substrate are then detectedand related to the presence of the DNA binding protein-DAN interations ofinterest in the sample. Also provided are devices and systems for use inpracticing the subject methods. The subject methods find use in a variety ofdifferent applications, e.g., detecting the presence of a transcription factorin a sample, the study of transcription factor profiles in response to a givenstimulus, screening for therapeutic agents, and the like.
