PURPOSE: Provided is a mass production of exo-polysaccharide from the submerged cultivation of Ganoderma lucidum by agitation and aeration effect under bi-staged pH controlling system of jar fermenter, thereby establishing scale-up conditions to maximize the production of exo-polysaccharide. CONSTITUTION: The method is characterized by the submerged cultivation of Ganoderma lucidum by stirring and aeration under bi-staged pH controlling system, wherein 0.1%(w/v) of polyacrylic acid is added to the submerged cultivation; stirring speed is 100-700 rpm and aeration rate is 0.3-2 vvm; and submerged cultivation is carried out at pH3, and 6-48 hours later, at pH6.
