METHOD FOR SIMULTANEOUSLY MEASURING ENZYME ACTIVITY OF 5amp;alpha;-REDUCTASE AND AROMATASE CYTOCHROME P450 WITH COMMON SUBSTRATE AND CO-ENZYME WITHIN A SHORT PERIOD OF TIME

PURPOSE: A method for simultaneously measuring the enzyme activity of 5α-reductase and aromatase cytochrome P450 is provided which is useful for biochemistry, pharmacy, toxicology and neurological, thereby simultaneously measuring two kinds of enzymes with a common substrate and a co-enzyme within a short period of time. CONSTITUTION: The method for simultaneously measuring the enzyme activity of 5α-reductase and aromatase cytochrome P450 comprises the steps of: (1) adding a buffer solution, testosterone as a common substrate, and NADPH as a common co-enzyme into a sample and reacting them at 37 deg. C for 20 minutes; (2) adding ethylacetate into the sample to terminate the reaction, and adding an inner standard material into the sample; (3) extracting the sample containing dihydrotestosterone and estradiol by centrifugation and evaporation of the sample; and (4) subjecting the extracted sample to GC(gas chromatography)/MS(mass spectrometry) to quantify the dihydrotestosterone and estradiol.
