PROBLEM TO BE SOLVED: To provide an autonomously replicable plasmid having a new replication system, and to provide an Escherichia coli-Rhodococcus bacterium shuttle vector.;SOLUTION: The autonomously replicable plasmid having the new replication system is, for example, obtained by confirming and selecting the presence or absence of the plasmid from Rhodococcus bacteria originated from the natural world and an allotting organization by an electrophoresis or the like. In order to obtain a plasmid having a replication system different from those of plasmids which have been known, the base sequence of the collected plasmid is compared with that of a replicated protein Rep A of a plasmid pAL 5000 originated from Mycobacterium fortuitum belonging to the family of ColE2 type plasmid.
