The present invention provides glycoimidazole (GIZ) and polyhydroxycyclohexenyl amine (PHCA) derivatives, methods for their manufacture, and methods of use wherein the GIZ and PHCA derivatives have a short flexible binder from the corresponding position of oxygen in the ring in a pyranose; and a lipophilic moiety attached to the binder and pharmaceutically acceptable salts thereof. More particularly, the present invention further relates to a method of treating individuals with Gaucher disease by administering the novel GIZ and PHCA derivatives as chaperones (linker proteins) for the disease-associated mutant glucocerebrosidase.
