A diagnostic kit and chip for gastric cancer are provided to maximize diagnosis accuracy and detect gastric cancer at an initial transformation step by detecting methylation of promoter CpG island by using a gastric cancer-specific methylation marker gene, thereby enabling early diagnosis. A diagnostic kit for gastric cancer comprises primers for amplifying a fragment containing CpG island of a gastric cancer marker gene promoter and endonuclease for digesting unmethylated nucleic acid sensitively, wherein the endonuclease is selected from SmaI, BssHI, HpaII, BSTUI, NotI, SacII and BagI, and the gastric cancer marker gene is screened by (a) comparing gene expressions of transformed cells with those of untransformed cells and confirming genes of which expressions are reduced in the transformed cells, (b) treating the transformed cells with a demethylation agent such as 5 aza 2'-deoxycytidine(DAC), comparing gene expressions of agent-treated transformed cells with those of agent-untreated transformed cells, and confirming genes of which expression are more increased in the agent-treated transformed cells, and (c) selecting the genes commonly confirmed in the steps(a) and (b) as gastric cancer maker genes.
