The sample chamber has a boundary region of a barrier layer (4), which limits a circular cell culture area (9)(9). Cell adhesion molecules (7b) of a fusion protein (7) act as platelet endothelial cell adhesion mollecule-1, junctional adhesion molecule, cadherin, integrin, occludin or claudin. The barrier layer consists of a corrosion resistant metal or plastic. A protein-binding layer (6) is applied between the region and the antibody fragments (7a) of the protein. A plate cone rheometer and a flat-parallel culture plate (1) are formed from glass and plexiglass and permit optical observations. The sample chamber in which electrodes are vacuum-metallized on the culture plate, which permits measurements of the transcellular electrical resistance of a cell culture medium. A ring plate holds the connections of the electrodes and leads to an electric coupling fastened at a ring insert. The sample chamber forms a gas-tight sample area, which includes an area of a shear slit between the culture plate and the cone. A capillary leads into the sample area for supply-or cooling gas-or active substance medium. The cone is fixed in a cone retainer and the culture plate is fixed on a culture plate carrier. A sealing gas-tightly seals the ring gap between the culture plate carrier and the cone retainer or the ring gap between the ring insert gas-tightly connected with the culture plate carrier and the cone retainer. The culture plate carrier is a single piece with the ring insert and is pinched over an easily solvable sealing-and fixing ring. The sealing is implemented as a soft-flexible lip-sealing ring fastened at the culture plate carrier and/or the ring insert. A temperature-controlled heating is integrated into the culture plate carrier and/or the ring insert for tempering of the sample area.
