A MOST(Media Oriented System Transport) interface apparatus having an emergency rescue request function, and a security system for a vehicle and an operating method thereof are provided to rescue a captured user safely from a vehicle trunk, by requesting the emergency rescue signal to related organizations. A MOST protocol processor(110) receives at least one of external signals, outputs received confirming signals, converts the external input signals to be adapted to the MOST protocol, and then transmits the converted signals to a multimedia device through a MOST network(120). A rescue signal input device(140) is installed within a vehicle trunk, and outputs the rescue signal if a key is inputted by a user. A receiver(150) receives the rescue signals. A controller(130) receives position information of the vehicle by requesting to a navigation(200) system, and outputs the rescue request signals periodically. A wireless transmitter(160) transmits the rescue request signal to external devices by converting the signals into RF signals.
