Exoribonuclease substrate, useful e.g. to detect and/or measure exoribonuclease activity in sample comprises first strand of nucleic acid which is RNA strand, and second strand of nucleic acid which is hybridized to first strand
Exoribonuclease substrate comprises a first strand of nucleic acid which is an RNA strand, and a second strand of nucleic acid which is hybridized to the first strand, where the substrate comprises, at one end, linking zone, which allows the exoribonuclease to be fixed and degrade the first strand and is marked with a part by a fluorescence emitter and the other by fluorescence silencer, one is fixed to the first strand and the other is on the second strand. Exoribonuclease substrate comprises a first strand of nucleic acid which is an RNA strand, and a second strand of nucleic acid which is hybridized to the first strand, where: the substrate comprises, at one end, linking zone, which allows the exoribonuclease to be fixed and degrade the first strand and is marked with a part by a fluorescence emitter and the other part by fluorescence silencer, one is fixed to the first strand and the other is on the second strand; and the emitter and silencer are fixed one against the other at an extinction distance of fluorescence emitted by the silencer, and does not prevent the activity of the exoribonuclease on the substrate. An independent claim is included for a method for analyzing the activity of a exoribonuclease in a sample, comprising: adding the substrate to the sample under conditions for allowing the exoribonuclease to be active if it is present in the sample; and measuring the fluorescence emitted by the fluorescence emitter, where the emission is caused by the degradation of the first strand by exoribonuclease, if it is present in the sample.
