The invention relates to immunochemical assay, namely to the quantitative determination method of phytohormones, 24S-methylbrassinosteroids – castasterone and brassinolide, and can be used for detection of these brassinosteroids in water solutions, quantitative estimate in biological objects, residual wastes in soil, manufacturing water et al. The task of the present invention is the quantitative determination method of 24S-methylbrassinosteroids (castasterone and brassinolide) and composition for its realization which is distinguished as highly sensitive, environmental safety, simplicity, reliability and manufacturability. The problem is solved, firstly, by claimed method consisted in immunochemical detection of 24S-methylbrassinosteroids using polyclonal antibodies for castasterone; secondly, by composition for claimed method realization which includes: antibodies for castasterone immobilized on polymer plates; castasterone-horseradish peroxidase conjugate; buffer solution; chromogen–substrate buffer (solution of 3,3',5,5'-tetramethylbenzidine in substrate buffer with hydrogen peroxide); stop reagent – 4.8% solution of H2SO4.
