The present invention relates to a process for the preparation of controlled drug formulation and method of coating comprising sirolimus which enhances stability and re-endothelialization. The invention also relates to a coated implantable medical device, comprising: a pharmacologically active core; a base coating comprising a hydrophilic polymer with silymarin; a drug layer comprising sirolimus and at least one surface modifying agent with a biodegradable polymer surrounding the said core; optionally, a barrier layer over the drug coated core containing silymarin.
