This invention relates to a process for simultaneous detection of two transgenes 5-enolpyruvylshikimate-3-phosphate synthase (cp4epsps) gene and CaMV 35S promoter in transgenic maize utilizing a combination of novel primer pair of sequence ID 1 and 2 for cp4epsps gene and of sequence ID 3 for CaMV 35S promoter in Multiplex PCR, Wherein each primer pair for amplifying 441 or 455 base pair of cp4epsps gene and 140 base pair of CaMV 35S promoter and the MPCR method comprising of initial denaturation at 94°C for 3 min followed by 40 cycles of 94°C for 30 sec, 60°C for 60 sec, 72°C for 60 sec, final reaction at 72°C for 5 min.
