PROCESS FOR PREPARING AN IGA1 PROTEASE FROM A CULTURE OF N. МЕNINGIТIDIS SEROGROUP A AND POLYVALENT VACCINE PREPARATION FOR THE PROPHYLAXIS OF AN INFECTION CAUSED BY BACTERIA OF N. МЕNINGIТIDIS SEROGROUPS А AND В

The process for preparing an IgA1 protease from N. meningitidis consists in cultivating a culture of N. meningitidis serogroup A (А 208 strain) on a modified Frantz medium, with subsequent inactivation of the culture liquid with cetavlon and precipitation of the microbial mass by means of centrifuging and isolation of the IgA1 protease by means of purification, which comprises ultrafiltration, absorption column chromatography on the sorbents silochrome 80 and phenyl-sepharose, and concentration and lyophilization of the product. Both fractions, namely the cetavlon supernatant and the cetavlon precipitate, are used. The resultant precipitate is extracted repeatedly, and the precipitate extracts and the supernatant are concentrated by ultrafiltration. The resultant concentrates are fractionated by absorption chromatography on a column with the sorbent silochrome 80, neutralized with 0.02 M of a phosphate buffered saline, wherein the IgA1 protease is prepared by washing the sorbent with this buffer solution. Using the IgA1 protease isolated from N. meningitidis, a polyvalent vaccine preparation is produced for the prophylaxis of an infection caused by bacteria of N. meningitidis serogroups А and В.
