The present invention relates to a novel culture medium, mycobacteria, in particular, the mycobacteria of the complex mycobacterium tuberculosis, making it possible to significantly reduce the time of the insulation by culture, and therefore the time of diagnosis of mycobacterioses, in particular of tuberculosis. A culture medium according to the invention contains blood defibrinated, lecithin, and of decomplemented foetal calf serum. The present invention also relates to a method of culture and identification of the mycobacteria, and in particular, bacteria of the complex mycobacterium tuberculosis. Still more particularly, the present invention relates to a novel process for the decontamination, by treatment with chlorhexidine, of the biological samples in a new isolating medium and culture of the mycobacteria. Finally, the present invention relates to a new identification method in the liquid phase by mass spectrometry, thus contributing to significantly reduce the time of diagnosis of mycobacterioses, in particular of tuberculosis.
