The invention relates to a method which includes: simultaneously lighting an object (O) using divergent light that includes three or more wavelengths (λ1 + λ2 + λ3 +...); recording a multiple hologram of the lit object (O); and obtaining, by digital reconstruction, a composite holographic image by digitally processing the multispectral information included in the multiple hologram relating to the three or more wavelengths (λ1 + λ2 + λ3 +...), which includes weighted mixing in the Fourier domain of the spectra corresponding to each wavelength. The microscope is suitable for carrying out the method from the first aspect. The computer program carries out the digital reconstruction of the method of the invention.
