The present invention relates to a composition and a sheet, comprising a mesenchymal stem cells-hydrogel-biodegradable support or a mesenchymal stem cells-hydrogel-non-degradable support, and a preparation method therefor. More specifically, the sheet comprising an adipose-derived mesenchymal stem cells-hydrogel-biodegradable or non-degradable support according to the present invention has beneficial effects of having excellent skin regeneration and wound treatment capabilities, and is capable of reducing treatment duration compared to conventional therapeutics because the stem cells having high activity can be applied to wound sites intactly without an isolation step using proteases, and an extracellular matrix, such as a collagen, a laminin, a fibronectin, and an elastin, secreted from the mesenchymal stem cells in a culture step is present to be intactly patched on a hydrogel.
