The present invention relates to an expression vector for establishing a high expression cell line comprising internal ribosomal entry site (IRES) and an antibiotic selecting marker gene expression cassette in which protein destabilization sequences are connected, a transformed cell using the high expression cell line, a method for establishing a high expression cell line, and a method for highly producing protein of interest (POI). The present invention is capable of efficiently producing recombinant protein by sorting only a high expression cell line rapidly and easily while blocking generation of false positive cells. An expression vector for establishing a high expression cell line according to the present invention comprises: (i) a gene expression cassette which encodes protein of interest (POI); (ii) a gene which encodes internal ribosomal entry site (IRES); and (iii) an antibiotic selecting marker gene expression cassette in which protein destabilization sequences are connected.;COPYRIGHT KIPO 2016
