The present invention relates to a method of inducing an immune response to CMV using a conditionally replicable deletion of a genetically modified cytomegalovirus (CMV). The methods of the present invention can be used to treat and / or prevent primary CMV infection, infection due to reactivation of latent CMV, and multiple infections caused by CMV strains other than those encountered previously. The invention also relates to a recombinantly altered replication defective CMV that allows viral replication to be externally controlled.
