A drug delivery system, comprising: a hydrogel formed from cyclodextrin and an amphiphilic triblock copolymer that includes a middle segment of block B polymer and two terminal segments of block A polymer, wherein the copolymer includes a block of polymer A comprising a poly (alkylene oxide) and a polymer block B comprising a poly (hydroxyalkanoate), and a therapeutically effective amount of at least one therapeutic agent intimately contained within the hydrogel; the poly (hydroxyalkanoate) block B polymer is selected from the group consisting of: poly [(R) -3-hydroxybutyrate]; poly [(R) -4-hydroxybutyrate]; poly [(R) -3-hydroxivalerate]; poly [(R) -3-hydroxybutyrate] -co-poly [(R) -3-hydroxivalerate]; poly [(R) -3-hydroxyhexanoate]; poly [(R) -3-hydroxyheptanoate]; enantiomers (S) of each of said enantiomers (R); racemic mixtures of said enantiomers (S) and (R); and mixtures.
