The invention provides: a method for enhancing transcription factor binding site-specific demethylation in DNA in a cell, the method including the overexpression, in a cell, of a gene that encodes a transcription factor that induces site-specific DNA demethylation; and a method for overexpressing a gene that encodes a transcription factor in a cell, identifying the demethylated site in the DNA in the cells, and screening the transcription factor that induces site-specific DNA demethylation, on the basis of the frequency of appearance of transcription factor binding motifs at the identified demethylation site and in the upstream sequence and downstream sequence thereof.
