The presently disclosed method for evaluating the biosafety of nano-zinc oxide based on 3D Caco-2 cells, which belongs to the technical field of biological determination or testing methods, comprises the following steps: 1) 3D cell culture; 2) transition; 3) dispersing nano-zinc oxide powder in DMEM culture medium without serum for later use; 4) observing nanomaterial characterization and cell morphology under 3D culture conditions, assaying cell activity by MTT colorimetric method, testing for inflammatory factors, interleukin-8 (IL-8), testing various cell death modes by flow cytometry; Perform analysis using SPSS19.0 statistical software, perform a one-way analysis on data, and calculate the significant difference of p 0.05 and p 0.01.
