The present invention relates to a composition including a primer specific to the TGLA-325 used as a microsatellite marker to identify one of the native dark brown cattle, Jeju black cattle, and Korean cattle, a kit including the composition for the same to identify one of the native dark brown cattle, Jeju black cattle, and Korean cattle, and a method using the composition or kit for the same to identify one of the native dark brown cattle, Jeju black cattle, and Korean cattle. According to the present invention, the microsatellite marker of four species are used to easily identify the native dark brown cattle, Jeju black cattle, or Korean cattle, thereby enabling various utilization in agricultural and food production fields through the systematic and scientific establishment of the genetic resources of livestock.
