The present invention relates to an immunity enhancing composition including the pituitary adenylate cyclase-activating peptide-38 (PACAP-38) as an active ingredient. As a result of executing a proteomic analysis, a hierarchical cluster analysis, and a signal transmission analysis on the plasma of mice exposed to smoking, the increased expression of PACAP-38 is confirmed in young mice while also confirming the anti-cancer effects of PACAP-38 caused by the increased cell killing capabilities of natural killer cells. The PACAP-38 with above-described effects can be utilized for pharmaceutical compositions, dietary supplement compositions, and anti-cancer compositions for immune enhancement as well as for immunosuppressant compositions.
