The present invention relates to the process of producing glucose from an OPEFB biomass, which consists of the following steps: a pretreatment of OPEFB with a method of NaOH base combined with steam explosion and a fed-batch enzymatic hydrolysis of the pretreated OPEFB. A pretreatment operation condition and enzymatic hydrolysis are optimized to obtain high cellulose conversion and glucose concentration with low NaOH and enzyme consumption, moderate pretreatment operating temperatures, and short pretreatment and enzymatic hydrolysis process time because these three factors will affect operating costs and investment costs. By using the optimum operation conditions obtained from the present invention, glucose concentration and conversion of cellulose to glucose are 170 g/L and 91%, respectively.
