A determining method and device for layer combination in a layered water injection process, and a machine-readable storage medium, relating to the technical field of oil and gas field development engineering. The method comprises: first, performing preset combination on water injection layers; then, aiming at "suppressing layers with less water breakthrough time and promoting layers with more water breakthrough time", on the basis of the water breakthrough time of each small layer in the water injection layers, determine the optimized injection volume for each small layer and calculating the variance of water breakthrough time and cumulative oil production at the water breakthrough time in each preset layer combination scheme; and finally, on the basis of the variance of water breakthrough time and cumulative oil production at the water breakthrough time, comprehensively evaluating each preset layer combination scheme. In the present determining method for layer combination, not only is the interlayer difference among the small layers considered, but also each preset layer combination scheme is further evaluated on the basis of the optimized injection volume for each small layer, so that the finally determined layer combination scheme can kept in line with oil well production conditions, and the purpose of optimizing layer combination can be achieved.
