A centerline extraction-based measurement method for providing a three-dimensional architecture of a plant root system, comprising the following steps: acquiring serial tomography image data of a plant root system; preprocessing the serial tomography image data of the plant root system, performing three-dimensional spatial filtering and expansion, and reconstructing a three-dimensional surface mesh model; performing iterative contraction on the three-dimensional surface mesh model to obtain a root system centerline; processing the root system centerline to obtain a centered centerline, reconstructing branch topological relations and performing smoothing processing to obtain a smoothed root system centerline; and performing, by means of the smoothed root system centerline, calculation with respect to root system parameters to obtain three-dimensional architectural parameters for the root system.
