摘要

Disclosed are a fusion primer for third-generation sequencing library construction, and a library construction method, a sequencing method and a library construction kit therefor. The fusion primer, from the 5' end to the 3' end thereof, sequentially comprises a homologous recombination sequence, a tag sequence and a specific amplification primer sequence, wherein the homologous recombination sequence is used for performing homologous recombination splicing on amplification products of the fusion primer; the tag sequence is used for distinguishing different amplification products; and the specific amplification primer sequence is used for binding to a target sequence in order to perform primer extension. By means of the amplification of the target sequence by using the fusion primer, and by means of performing homologous recombination splicing on the amplification products, the number of samples on a sequencing chip is increased and the cost of sequencing is reduced.
