The present invention relates to a primer set for reverse transcription polymerase chain reaction (RT-PCR) for multi-diagnosis on virus and viroid infection generated from an apple or a pear; a composition; a diagnosing kit; and a diagnosing method. The present invention relates to a multi-diagnosing primer set composed of a combination of primer sets capable of effectively diagnosing Apple mosaic virus (ApMV), Apple stem grooving virus (ASGV), Apple stem pitting virus (ASPV), Apple chlorotic leaf spot virus (ACLSV), and Apple scar skin viroid (ASSVd) at the same time; a composition; a diagnosing kit; and a diagnosing method. When conducting RT-PCR, the diagnosing composition helps to quickly and conveniently diagnose viruses and viroids of only an apple or a pear from an infection sample without false positive reaction, thereby providing effects of reduction in diagnosis costs and efforts. Therefore, the primer set for RT-PCR and the diagnosing method thereof can solve problems in agricultural sites by effectively diagnosing fruit tree viruses and viroids and thus, producing and distributing high-quality apple or pear seedlings.
