Development of Inactivated Vaccines for Parainfluenza Virus. Annual Report September 30, 1982-September 29, 1983

Most of the authors effort has been placed on large scale preparation of highly purified parainfluenza virus-3 glycoproteins. The optimal conditions for virus growth have been reexamined including the cell type, growth conditions, multiplicity of infection and the time of harvest. Optimal conditions for the detergent extractions of the viral glycoproteins have been established. In preliminary studies, the two viral glycoproteins HN and F have been separated successfully by cation exchange chromatography on carboxymethyl cellulose (CM-52) columns from detergent extracted viral envelope preparations. Conditions for radioimmune precipitation for the identification of antibodies to different viral proteins have been established. The authors are also continuing to develop monoclonal antibodies to (PI-3) viral gycloproteins.
