The National Toxicology Program (NTP) has recently developed and is evaluating a new reproductive toxicity system. This test system is designated 'Fertility Assessment by Continuous Breeding' (FACB). Caesarean originated Barrier-Sustained COBS CD-1, (ICR)BR outbred albino mice are used for the FACB study. It consists of four related tasks, not all of which are necessarily performed for a given compound (Figure 1). These tasks include, Task 1, dose finding; Task 2, continuous breeding phase; Task 3, identification of the affected sex; and Task 4, offspring assessment. This test protocol is designed to provide both time and cost effective alternatives to multigeneration studies which produce similar comprehensive reproductive data. In order to evaluate the FACB protocol, the NTP is testing a variety of chemicals in two different laboratories. This report describes the effect(s) of lead acetate trihydrate on male and female reproduction when tested according to the FACB protocol at Environmental Health Research and Testing, Inc., Lexington, Kentucky.
