Single pellets, each containing 3 mg of 8-Cl-cAMP (NSC-614491) and 17 mg of cholesterol, were implanted subcutaneously in male CD2F1 mice. Blood samples from 2 or 3 mice/time point were collected at various times from 1 to 96 hours after pellet placement. Plasma from these samples was analyzed for 8-Cl-cAMP concentration. Maximum levels of approximately 4-9 micrograms/mL were noted at 1 hour, the earliest time point examined. Minimal levels (0.1-0.2 micrograms/mL) were noted after 6 hours. Levels declined rapidly and approached the limit of detection by 16 hours in the first study and 56 hours in the second study. Examination of the implantation sites at 96 hours after placement showed that the pellets remained unchanged in physical appearance.
